The Silent Information Regulator (SIR2) family of genes are highly conserved from prokaryotes to eukaryotes and have important functions in the regulation of metabolism, growth and differentiation, inflammation, cellular survival, as well as in senescence and lifespan extension. Sirtuins, including SIRT1-7, are human homologs of yeast Sir2p. Sirtuins are NAD+-dependent histone/protein deacetylases (HDAC) which regulate cellular metabolism, e.g. energy metabolism, and thereby are associated with aging and several age-related diseases. SIRT4 localizes to mitochondria, inhibits glutamate dehydrogenase, and is thought to be involved in the regulation of insulin secretion. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: ELISA: 1:128,000 Western Blot: Detects a band at ~37kD in human kidney and U937 lysates Immunohistochemistry (Paraffin): 2-3ug/ml. Antigen retrieval with citrate buffer, pH 6 is required. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
