Bacterially produced recombinant protein corresponding to human SIT, intracellular fragment.
SIT (SHP2-interacting transmembrane adaptor protein) is expressed exclusively in lymphoid organs and acts either as a positive or as a negative regulatory element in T cell activation and in T cell development. Binding to Grb2 plays a pivotal role in signal transduction. Hubener et al. (2001) determined that the SIT gene contains 5 exons and spans 1.8 kb of genomic DNA. The SIT promoter demonstrated strong transcriptional activity and potential binding sites for both ubiquitous and lymphoid-specific transcription factors. Applications: Suitable for use in Immunoprecipitation, Flow Cytometry and Western Blot. Other applications not tested. Recommended Dilution: Flow Cytometry: 1-5ug/ml with intracellular staining. Western Blot: 1-2ug/ml under reducing conditions. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
