Siglec-1 cell surface antigen, is a 185kD molecule expressed by macrophages, predominately confined to lymphoid organs only. Monocytes and granulocytes are negative. No other cell types are positive. The most conspicious property of Siglec-1 is it stains marginal zone macrophages and marginal metallophils in the spleen very strongly. Furthermore, macrophages in (auto-immune) diseased tissues express the Siglec-1 antigen. In healthy tissue no expression occurs. Siglec-1 is a receptor for glycoconjugates containing sialic acid. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunofluorescence and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. Hybridoma: Sp2/0-Ag 14 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
