Siglec-1 cell surface antigen, is a 185kD molecule expressed by macrophages, predominately confined to lymphoid organs only. Monocytes and granulocytes are negative. No other cell types are positive. The most conspicious property of Siglec-1 is it stains marginal zone macrophages and marginal metallophils in the spleen very strongly. Furthermore, macrophages in (auto-immune) diseased tissues express the Siglec-1 antigen. In healthy tissue no expression occurs. Siglec-1 is a receptor for glycoconjugates containing sialic acid. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunofluorescence and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. Hybridoma: Sp2/0-Ag 14 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
