Synthetic peptide corresponding to the N-terminal sequence of human Survivin.
Survivin is a 16kD antiapoptotic protein highly expressed during fetal development and cancer cell malignancy (1). By binding and inhibition of caspase-3, Survivin controls the checkpoint in the G2/M-phase of the cell cycle to inhibit apoptosis and promote cell division (2,3). This regulation process requires the phosphorylation of Survivin at Thr34 by p34cdc2 kinase (4). Gene targeting using the Thr34 phosphorylation defective Survivin mutant as well as antisense Survivin has been shown to inhibit tumor growth (5,6). Applilcations: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% nonfat dry milk, 1X TBS, 0.1% Tween-20 at 4C with gentle shaking, overnight. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
