Paired helical filaments (PHF) tau preparation from human brain.
Alzheimers disease (AD) is characterized by senile plaques, which are composed of extracellular deposits of Ab peptide, and neurofibrillary (NF) tangles. The NF tangles are composed of intracellular filamentous aggregates of hyperphosphorylated tau proteins. NF tangles consist of microtubule-associated protein of paired helical filaments (PHF-t) that are biochemically and structurally distinct from the amyloid fibrils in senile plaques. Clone PHF-6 recognizes phosphorylation dependent epitopes of Thr-231 on PHF-t as well as fetal tau, but does not recognize autopsy-derived normal adult tau. Phosphoamino Thr-231 is one of the two major immunodominant residues of PHF-t. The antigen recognition pattern of PHF-6 is completely identical to that of monoclonal AT180 (widely used clone). PHF-6 does not recognize unphosphorylated or Ser235 phosphorylated 224-240 peptide, and it recognizes the Thr231 phosphorylated and diphosphorylated peptides equally well. Applications: Suitable for use in ELISA and Western Blot. Other applications have not been tested. Recommended Dilution: ELISA: 0.1-1ug/ml Western Blot: 1-3ug/ml Optimal dilutions to be determined by the researcher. Positive Control: Human fetal brain preparation and with biopsy derived, human paired helical filament preparation from brain. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
