TCR affinity purified from mouse T-cell hybridoma DO-11.10
The alpha/beta TCR is expressed on T lymphocytes of all mouse strains tested.2 Plate-bound or soluble antibody activates a/b TCR-bearing T cells.2 In vitro and in vivo, this antibody can induce immature thymocytes to undergo apoptosis.3,4 In vivo administration of this antibody has been shown to deplete a/b TCR-bearing cells to near completion and prevent graft rejection.5 Applications: Flow cytometry 2,6,7 Immunohistochemistry (acetone-fixed, frozen sections and formalin-fixed, paraffin-embedded sections) Immunoprecipitation 2,8 In vitro stimulation of a/b TCR-expressing T cells 2 In vitro and in vivo depletion of a/b TCR-bearing cells 9 Recommended Dilution: Flow Cytometry: 0.2ug/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20C or colder. Aliquots are stable for at least 6 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. PE/CY5 combination conjugate is used as a third fluorochrome in multicolor flow cytometry applications when only a single 488nm excitation source is available. Antibodies are covalently coupled using heterobifunctional linking reagents, resulting in an optimal conjugation format for flow cytometry.
