Tetanus toxin, a member of the peptidase M27 family of proteins, is produced by the anaerobic spore-forming bacteria Clostridium tetani as a single-chain polypeptide that is 1315aa in length. The protein is subsequently cleaved by an endogenous protease to yield the 52kD tetanus toxin light chain (TeNT-LC), which is 457aa in length, and the 98kD tetanus toxin heavy chain that is 858aa long. The light and heavy chains are linked by a disulfide bridge and are non-toxic after separation. Tetanus toxin acts by inhibiting neurotransmitter release. It binds to peripheral neural synapses, is internalized and moves by retrograde transport up the axon into the spinal cord where it can move between postsynaptic and presynaptic neurons. It inhibits neurotransmitter release by acting as a zinc endopeptidase that catalyzes the hydrolysis of the 76-Gln-|-Phe-77 bond of synaptobrevin-2. Applications: Suitable for use in FLISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
