Immunogen: Synthetic peptide corresponding to the N-terminal region of human TIMP-2.
TIMP-2 is closely related to TIMP-1 and shows the highest binding affinity to both the latent (pro) and active forms of 72kD Type IV collagenase (also known as MMP-2 or gelatinase A). It also has affinity for the active form of 92kD Type IV collagenase (also known as MMP-9 or gelatinase B). TIMP-2 innibits the proteolytic invasiveness of tumor cells and normal placental trophoblast cells. May be used to evaluate the invasive nature of some tumor types and determine the tissue distribution of TIMPs in other tumore such as adenomas and also its normal tissue. Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (formalin/paraffin only): 2-4ug/ml 30 min at RT. Western Blot: 1ug/ml for 2 hrs at RT. Note: Staining of formalin-fixed tissues REQUIRES boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min. followed by cooling at RT for 20 min Optimal dilutions to be determined by the researcher. Positive Control: Pancreatic or colon carcinoma Cellular Localization: Cytoplasmic Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
