The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules. The TLR family is a phylogenetically conserved mediator of innate immunity that is essential for microbial recognition. Ten human homologs of TLRs (TLR1-10) have been described. Among this family of receptors, TLR2 and TLR4 have been most studied. These studies have suggested that TLR2 and TLR4 may serve as potential main mediators of LPS signaling. The TLR4 cDNA codes for a protein consisting of 799 amino acids with approximate molecular weight of 88kD. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 2-5ug per 10e6 cells Immunohistochemistry: Paraffin Western Blot: 1-3ug/ml Optimal dilutions to be determined by the researcher. Positive Controls: Flow Cytometry: Ramos, ThP1 Western Blot: Intestine lysate Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
