MaxLight(TM)490 is a new Blue-Green photostable dye conjugate comparable to DyLight(TM)488, Alexa Fluor(TM)488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm), Emission (515nm), Extinction Coefficient 73,000. TLR4 has been demonstrated to act as a receptor for LPS on human monocytes and macrophages. TLR4 signalling of LPS stimulation requires the presence of the MD-2 molecule. TLR4 is weakly expressed by resting cells, but is upregulated following stimulation with LPS. This antibody has been demonstrated to block activation of monocytes with LPS. Removal of sodium azide is recommended prior to use in functional assays. Applications: Suitable for use in Immunoprecipitation and Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:5-1:12.5, Use 10ul of the suggested working dilution to label 10e6 cells or 100ul whole blood. Optimal working dilutions to be determined by researcher. Hybridoma: Sp2/0 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
