Antigen-specific cell contacts in the immune system are strengthened by antigen-nonspecific interactions, mediated in part by lymphocyte-function associated (LFA) antigens. Recently, CD54 (intercellular adhesion molecule-1) has been defined as a ligand for LFA-1. Monoclonal antibodies to CD54 block T lymphocyte adhesion to fibroblasts and endothelial cells and disrupt the interaction between cytotoxic T cells and target cells. CD54 is found on leukocytes, fibroblasts, epithelial cells and endothelial cells and its expression is regulated by inflammatory cytokines. The normal function of human CD54 is to provide adhesion between endothelial cells and leukocytes after injury or stress. However, CD54 is also used as a receptor by the major group of human rhinoviruses and is a catalyst for the subsequent viral uncoating during cell entry. Monoclonal antibodies recognize a 95kD cell surface glycoprotein the major human rhinovirus receptor, CD54. Applications: Suitable for use in ELISA, Western Blot, Immunocytochemistry, Immunohistochemistry and Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 2ug/ml Western Blot: non-reducing conditions. Immunohistochemistry (paraffin sections): 10ug/ml, positive tissue: thymus, RE cells. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
