MaxLight(TM)490 is a new Blue-Green photostable dye conjugate comparable to DyLight(TM)488, Alexa Fluor(TM)488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm), Emission (515nm), Extinction Coefficient 73,000. Siglec-1 cell surface antigen, is a 185kD molecule expressed by macrophages, predominately confined to lymphoid organs only. Monocytes and granulocytes are negative. No other cell types are positive. The most conspicious property of Siglec-1 is it stains marginal zone macrophages and marginal metallophils in the spleen very strongly. Furthermore, macrophages in (auto-immune) diseased tissues express the Siglec-1 antigen. In healthy tissue no expression occurs. Siglec-1 is a receptor for glycoconjugates containing sialic acid. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunofluorescence and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. Hybridoma: Sp2/0-Ag 14 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight(TM)490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
