Synphilin 1 is a cytoplasmic protein that is enriched in the neurophils of normal human brain. Although the exact function of synphilin 1 is unknown, it may possibly have a role as a cytoskeletal protein based on the presence of ankyrin-like repeats and an ATP, GTP-binding domain. It has been shown to be novel is its ability to associate in vivo with alpha-synuclein to form cytoplasmic eosinophilic inclusions. Additionally, it has been observed to be present in Lewy bodies of Parkinson disease, which are composed of alpha-synuclein. Because synphilin 1 is not found in protein aggregates of Alzheimers disease, it appears to be specific for alpha-synuclein-containing lesions. Synphilin 1 may facilitate the aggregation of alpha-synuclein into Lewy bodies. Applications: Suitable for use in Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry: 1:1000-1:2000. Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
