T0710-05X defines the mucin-carried sialylated-Tn epitope. Tumor-associated glycoprotein (TAG-72) is an oncofetal antigen with properties of a mucin. About 60% of carcinoma patients express TAG-72 in their sera. A majority of human adeno-carcinomas including colorectal, pancreatic, gastric, ovarian, endometrial, mammary, and non-small cell lung cancer display some cell populations that are positive for TAG-72 staining. Weak or no reactivity has been observed with most cell types of normal adult tissue, except the secretory endometrium. It is reportedly useful in distinguishing pulmonary adenocarcinomas from pleural mesotheliomas. Applications: Suitable for use in Flow Cytometry, Radioimaging and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry (Formalin/paraffin): 1:500 for 20 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 20 minutes followed by cooling at RT for 20 minutes. Optimal dilutions to be determined by the researcher. Positive Control: Lung or colon carcinoma Storage and Stability: May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
