Talin-1 is a high molecular weight cytoskeletal protein, linking vinculin to the integrins, thereby linking the cytoskeleton to the extracellular matrix. It is specifically found in regions of cell-substratum contact and in lymphocytes at cell-cell contact points. Talin-1 plays a role in the assembly of actin filaments and in the migration of cells such as osteoclasts and fibroblasts. Applications: Suitable for use in Immunohistochemistry, Immunoprecipitation, Immunofluorescence and Western Blot. Other applications have not been tested. Recommended Dilutions: Immunohistochemistry (paraffin): 1:125-1:200 Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Immunofluorescence: Requires fixation and permeabilisation using methanol. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
