Immunogen: Synthetic phosphopeptide derived from the region of human tau that contains serine 262.
Tau is a neuronal microtubule- associated protein found predominantly on axons. The function of tau is to promote tubulin polymerization and stabilize microtubules. Tau, in its hyper-phosphorylated form, is the major component of paired helical filaments (PHF), the building block of neurofibrillary lesions in Alzheimers disease (AD) brain. Hyper-phosphorylated tau is also found in neurofibrillary lesions in a range of other central nervous system disorders. Hyper-phosphorylation impairs the microtubule binding function of tau, resulting in the destabilization of microtubules in AD brains, ultimately leading to the degeneration of the affected neurons. Numerous serine/threonine kinases, including GSK-3beta, protein kinase A (PKA), cyclin-dependent kinase 5 (cdk5) and casein kinase II, phosphorylate tau. Serine 262 can be phosphorylated by GSK-3beta, PKA, CamKII and MARK and has been found to be a major site in AD brain. This antibody, combined with Tau [pS356]), enables one to distinguish between phosphorylation at the two distinct residues, rather than a combined epitope as detected by 12E8 antibody. Applications: Suitable for use in Western blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Positive Control Used: Recombinant human Tau or African green monkey kidney (CV-1) cells, stably expressing human four repeat tau and SV40 small T antigen to induce specific inhibition of PP2A. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
