TCCR, also known as WSX-1 and IL-27 Ra, is a 96kD member of the type I group 2 cytokine receptor family. The extracellular region of human TCCR shares 63% aa identity with the extracellular domain of mouse IL-27 Ra. TCCR is expressed in mast cells, endothelial cells, NK cells, macrophages, monocytes, B cells, dendritic cells, and native T cells. As with many other class I cytokine receptor chains, IL-27 Ra heterodimerizes with gp130, the common receptor subunit of IL-6 family cytokines, to form a functional IL-27 receptor. IL-27 only binds to IL-27 Ra/WSX-1/TCCR and not the gp130 subunit. Studies using WSX-1 mice reveal that WSX-1 has the ability to suppress T cell hyperactivity during infection, and to mediate an inhibition of both type 1 and type 2 T cell immunity. In particular, WSX-1 is believed to induce IL-12 R on native T cells, predisposing them to a Th1 phenotype. This is offset by its ability to limit IL-12 (and TNF) production by macrophages, and its suppression of IFN-g production. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 human peripheral blood mononuclear cell (PBMC) lymphocytes Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Cells may be Fc-blocked with 1ug of human IgG/10e5 cells for 15 min at RT Do not wash excess blocking IgG from this reaction. 2. After blocking, 10ul of T2009-04B was added to up to 1x10e6 cells and incubated for 30 min at RT. 3. Unbound antibody was removed by washing the cells twice in F4568-10: Flow Cytometry Staining Buffer, 1X. Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Human Lyse Buffer. 4. The cells were resuspended in F4568-10 for final flow cytometric analysis. As a control for this analysis, cells in a separate tube should be treated with a mouse IgG 2b (PE) antibody. Storage and Stability: Store product at 4C in the dark. DO NOT FREEZE! Stable at 4C for 12 months after receipt as an undiluted liquid. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
