Immunogen: Human HA1.7 TCR beta chain expressed on transgenic mouse cells.
Human T cell receptor (TCR) is a T cell surface expressed heterodimer of a/b or g/d chains. TCRs recognize antigen peptides bound to MHC molecules forming the basis of antigen specific response by T cells. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50, 80ul labels 5x10e5 human Jurkat cells. Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 human Peripheral Blood mononuclear cells were washed and preincubated 5 minutes with 20ul human IgG at 250ug/ml. Cells were then incubated 45 minutes with 80ul T2040-04A at 1:50 (10ug/ml). 2. Cells were then washed three times, fixed and analyzed by FACS using a lymphoid gate. 3. A net 37% sub population of the cells stained positive with a mean shift of 1.26 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control at a similar concentration. 4. Binding was blocked when cells were pre-incubated with 20ul of 0.5mg/ml T2040-04. Storage and Stability: Store product at 4C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20C. Aliquots are stable at -20C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
