TIE2, phosphorylated (pTyr992) (Tunica Internal Endothelial Cell Kinase, Angiopoietin1 Receptor, p140 TEK, Tyrosine Protein Kinase Receptor) is a receptor tyrosine kinase (RTK). It is expressed almost exclusively on endothelial cells. It is critical for vasculogenesis during development and could be important for maintaining endothelial cell survival and integrity in adult blood vessels as well as tumor angiogenesis (1-3). The ligand for Tie has yet to be identified. A family of ligands known as the angiopoietins have recently been shown to bind Tie2. These ligands appear to have opposing actions as Angiopoietin-1 Ang1) and Angiopoietin-4 (Ang4) stimulate tyrosine phosphorylation of Tie2. Angiopoietin-2 (Ang2) and Angiopoietin-3 (Ang3) can inhibit this phosphorylation (4, 5). The phosphorylated Tie2 associates with different downstream signaling components. Grb2, Grb7, Grb14, Shp2, the p85 subunit of phosphatidylinositol 3-kinase and p56Dok-2 can interact with TIE2 in a phosphotyrosine- dependent manner through their SH2 or PTB domains (6, 7). Tyrosine 992 is located on the putative activation loop of Tie2 and is a major autophosphorylation site (8). Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
