Embryonic vascular system undergoes a series of complex, highly regulated series of events involving differentiation, migration and association of primitive endothelial cells. This process is termed vasculogenesis. A further remodeling of the primitive vascular system forms the mature cardiovascular system. This process is known as angiogenesis (sprouting of new capillary vessels from pre-existing vasculature). A family of receptor tyrosine kinases TIE1 and TIE 2 or Tek has been identified in vascular endothelium and hematopoietic cells. Mice lacking TIE 1 or TIE 2 are lethal. Ties may represent the earliest endothelial cell lineage marker and may regulate the endothelial cell proliferation, differentiation, and proper patterning during vasculogenesis. TIEs appear to be acting downstream of the VEGFRs. Tie-2 (human 1124aa, mouse 1122aa) is a type 1 membrane receptor protein specifically expressed in developing vascular endothelial cells and their progenitors, angioblasts. It is also found in placenta and lung, with lower levels in umbilical vein endothelial cells, brain and kidney. Tie-2 extracellular portion (25-279aa) contains 3 fibronectin type III-like and 2 Ig- like C2-type, and 3-EGF-like domains. Applications: Suitable for use in Western Blot, ELISA, and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1-10ug/ml (Chemiluminescence) ELISA: 1:10,000-1:100,000, 50-100ng of control peptide/well Immunohistochemistry: 2-20ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile ddH2O or PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
