Urea is the major end product of nitrogen metabolism and it is concentrated up to four times higher than that of plasma. A rapid, passive transport of urea through the membranes of the IMCD is responsible for generation of high urea concentration in the inner medullary interstitium. Recently, a vasopressin-sensitive urea transporter (UT2, 397aa) from rabbit and its homolog from rat (rUT2, 397aa) have been cloned. Applications: Suitable for use in ELISA, Western Blotting. Other applications not tested. Recommended Dilution: Western Blotting : (1:1000-1:5000 for neat serum and 1-10ug/ml for affinity pure antibody using ECL. An antibody made to the rUT2 peptide detected a major band at ~97kD in rat renal inner medulla membranes (2). The open reading frames of the cDNA clones for rabbit and rat UT2 predicted a protein of only ~43kD (1-3). However, northern blots using these cDNAs revealed 2 distinct transcripts (2.9 and 4 kb) (4). The 97kDband correspond to the 4 kb transcript (2). ELISA: 1:10,000-50,000 for neat serum and 0.5-1ug/ml for affinity pure. Control peptide can be used to coat ELISA plates at 1ug/ml. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.. We recommend the use of 0.5-1% milk in all primary/secondary antibody-enzyme conjugate incubations in order to suppress non-specific bands.
